Biological Techniques is a series of volumes aimed at introducing to a wide audience the latest advances in methodology. The pitfalls and problems of new techniques are given due consideration, as are those small but vital details not always explicit in the methods sections of journal papers. In recent years, most biological laboratories have been invaded by computers and a wealth of new DNA technology and this will be reflected in many of the titles appearing in the series. The books will be of value to advanced researchers and graduate students seeking to learn and apply new techniques, and will be useful to teachers of advanced undergraduate courses involving practical or project work. Methods of constructing artificial membranes (planar lipid bilayers) from the main components of cell membranes (lipids) date from the early 1960s. Planar bilayers offer direct, quantitative experimental approaches to the study of membranes of precisely determined composition which can be manipulated by the experimenter. Pore-forming molecules, transporter molecules, ATP-dependent enzymes and other entities can be incorporated into the bilayers to simulate biological functions. Reconstitution of such functions in this way remains a key final step in attributing a functional role to purified cell membrane proteins. This book aims to demystify these techniques and begins with a broad overview of the development of the subject before dealing with the protocols involved. Key references are provided at the end of the book together with a list of suppliers. Full practical details include: How to set up conventional painted and folded bilayer experiments Patch-dipping methods and use of giant liposomes Lipid characterization, preparation and purification Discussion of construction of essential apparatus, flux measurements, electrical recording, data acquisition and computer support Biochemical methods for use in planar bilayer experiments Techniques for incorporation of native proteins and other molecules

This book summarizes the current status of research on bilayer lipid membranes (planar lipid bilayers and spherical liposomes). In addition to describing the properties of lipid bilayers and examining biomembrane phenomena, the book has two other objectives. The first is to present practical methods for the formation and study of lipid bilayers with either aqueous or metal-lipid bilayer interfaces. The second aim is to treat planar lipid bilayers as a new type of interfacial adsorption phenomena. The first nine chapters cover properties of biomembranes, basic principles of membrane biophysics, transport, electrochemistry, physiology, bioenergetics, and photobiology. Chapter 10 presents the following topics: lipid bilayers in medicine, supported lipid bilayers as sensors, a short discussion of liposomes, and solar energy transduction via semiconductor septum photovoltaic cells based on natural photosynthesis.

Advances in Lipid Research, Volume 21 covers several significant and emerging areas in lipid metabolism, including apolipoprotein metabolism and leukotrienes. This book discusses the role of fat in the functioning of the immune system. Organized into eight chapters, this volume begins with an overview of the metabolism of apolipoprotein B, which is the principal protein of chylomicrons, very low density, and low density lipoprotein. This text then describes the metabolism, biosynthesis, and pharmacology of leukotrienes. Other chapters consider the relation of lipids to immunology and explain the roles played by lipids in the structure and function of yeast membrane. This book discusses as well the general importance in the plant kingdom and the place of some carotenoids in human biology. The final chapter deals with the influence of proteins on configuration and function of reconstituted lipid membranes. This book is a valuable resource for biologists, chemists, biochemists, scientists, and research workers.

In Bilayer Lipid Membranes. Structure and Mechanical Properties the authors use new methods of measurement, which they have themselves developed, to present an analysis of the relation between membrane structure and viscoelastic properties, in particular in the transversal direction. Hianik and Passechnik's approach is fundamentally different from the usual one, in that they analyze lipid bilayer dynamics during various modes of deformation, arriving at a new, `three-layer' model that accounts for the great heterogeneity of biomembranes. The macroscopic parameters of membranes have been measured using a wide variety of methods, leading to a discussion of the correlations between the parameters. There is also an extensive discussion of the dynamic changes in mechanical properties of lipid bilayers in the course of conformational transition of integral proteins. During the conformational changes of proteins, the structure of a bilayer undergoes a transition, reaching a new, stable membrane state. The book is the first to present a comprehensive analysis of long-distance interaction in lipid bilayers and of molecular mechanisms of mechanoreception. Audience: Scientists and graduate students working in biophysics, membranology, physiology, medicine, pharmacology, bioelectronics, electrochemistry, and colloid chemistry.

From the Introduction: Nanotechnology and its underpinning sciences are progressing with unprecedented rapidity. With technical advances in a variety of nanoscale fabrication and manipulation technologies, the whole topical area is maturing into a vibrant field that is generating new scientific research and a burgeoning range of commercial applications, with an annual market already at the trillion dollar threshold. The means of fabricating and controlling matter on the nanoscale afford striking and unprecedented opportunities to exploit a variety of exotic phenomena such as quantum, nanophotonic and nanoelectromechanical effects. Moreover, researchers are elucidating new perspectives on the electronic and optical properties of matter because of the way that nanoscale materials bridge the disparate theories describing molecules and bulk matter. Surface phenomena also gain a greatly increased significance; even the well-known link between chemical reactivity and surface-to-volume ratio becomes a major determinant of physical properties, when it operates over nanoscale dimensions. Against this background, this comprehensive work is designed to address the need for a dynamic, authoritative and readily accessible source of information, capturing the full breadth of the subject. Its six volumes, covering a broad spectrum of disciplines including material sciences, chemistry, physics and life sciences, have been written and edited by an outstanding team of international experts. Addressing an extensive, cross-disciplinary audience, each chapter aims to cover key developments in a scholarly, readable and critical style, providing an indispensible first point of entry to the literature for scientists and technologists from interdisciplinary fields. The work focuses on the major classes of nanomaterials in terms of their synthesis, structure and applications, reviewing nanomaterials and their respective technologies in well-structured and comprehensive articles with extensive cross-references. It has been a constant surprise and delight to have found, amongst the rapidly escalating number who work in nanoscience and technology, so many highly esteemed authors willing to contribute. Sharing our anticipation of a major addition to the literature, they have also captured the excitement of the field itself in each carefully crafted chapter. Along with our painstaking and meticulous volume editors, full credit for the success of this enterprise must go to these individuals, together with our thanks for (largely) adhering to the given deadlines. Lastly, we record our sincere thanks and appreciation for the skills and professionalism of the numerous Elsevier staff who have been involved in this project, notably Fiona Geraghty, Megan Palmer and Greg Harris, and especially Donna De Weerd-Wilson who has steered it through from its inception. We have greatly enjoyed working with them all, as we have with each other.
